Photosynthesis measurements are vital for understanding plant physiology and the environment. Advancements in these measurement techniques can help faculty and students study the connections between plants and other organisms, including their importance and function within ecosystems around the world.
Through the LEEF program, faculty members can acquire up to five different plant physiology packages.
Three packages feature the LI-6800 Portable Photosynthesis System: the LI-6800 LEEF Photosynthesis Package, the LI-6800 LEEF Gas Exchange and Fluorescence Package, and the LI-6800 LEEF Ecophysiology Package. Two packages feature the LI-600 Porometer/Fluorometer: the LI-600P LEEF Porometer Package and the LI-600PF LEEF Porometer/Fluorometer Package.
Paired together, the LI-6800 and LI-600 provide students with in-depth and survey analysis capabilities. The LEEF program also provides a grant that partially covers the cost of each package for any eligible institution.
Each LEEF plant physiology package can help students answer many fundamental questions related to ecophysiology. Faculty can use these packages to help students explore numerous aspects of plant stress, photosynthetic carbon assimilation, and chlorophyll a fluorescence.
The LI-6800 Portable Photosynthesis System featured in three LEEF packages has automated control over chamber conditions—such as leaf chamber temperature and H2O. It features high-precision H2O and CO2 analyzers mounted in the sensor head, close to the leaf. This allows for leaf-level feedback control. Changes in leaf dynamics are measured in real time. This helps students explore the effects of changing a single variable while holding others constant. Additionally, it has a user-friendly touch screen interface that facilitates the learning of photosynthetic responses.
The LI-6800 LEEF Photosynthesis Package includes a clear top chamber and light source. This package can be expanded at any time with the addition of a fluorometer.
The LI-6800 LEEF Gas Exchange and Fluorescence Package includes the Multiphase Flash™ Fluorometer. In addition to basic gas exchange measurements, this fluorometer enables simultaneous chlorophyll fluorescence and gas exchange measurements over the same leaf area in order to get more information regarding the dynamic leaf-level processes of photosynthesis.
The LI-6800 LEEF Ecophysiology Package expands system capabilities with a leaf chamber fluorometer and interchangeable chambers. These chambers are designed for exploring unique research opportunities—such as leaf chlorophyll fluorescence, soil respiration, and insect respiration.
The LI-600 Porometer/Fluorometer is a lightweight and handheld porometer and Pulse Amplitude Modulation (PAM) fluorometer device that can take a measurement in seconds. It delivers accurate, high-throughput measurements over the same leaf area. The compact, one-handed design makes it quick and convenient to take stomatal conductance and chlorophyll a fluorescence measurements—enabling students to analyze more plants in less time.
The LI-600P LEEF Porometer Package includes the LI-600P Porometer; this model allows students to take stomatal conductance measurements. Stomatal conductance to water (gsw) is a measure of the degree of stomatal openness and density. This measurement helps students understand a plant’s genetic composition and physiological response to its environmental conditions.
The LI-600PF LEEF Porometer/Fluorometer Package includes the LI-600PF Porometer/Fluorometer; this model allows students to take stomatal conductance and chlorophyll a fluorescence measurements. Chlorophyll a fluorescence measurements help students understand a leaf’s quantum efficiency, electron transport rate (ETR), and non-photochemical quenching (NPQ)—along with other reactions that collectively protect a leaf when it absorbs excessive light energy. Additionally, combined stomatal conductance and chlorophyll a fluorescence measurements present them with a more complete picture of a plant’s physiological state than either technique could provide on its own.
The LEEF program’s plant physiology packages are designed to help students address ecophysiological questions. They are a valuable teaching aid for faculty members when discussing photosynthesis and respiration both inside and outside the classroom.
The LI-6800 and the LI-600 work together to provide students with survey and in-depth measurements. They can screen plants with the LI-600 first to establish stomatal conductance and chlorophyll a fluorescence; both measurements will help pinpoint which plants to study in detail with the LI-6800. The LI-6800 environmental controls allow students to maintain parameters—from leaf temperature and light conditions to carbon dioxide and water vapor deficit—that can help answer their research questions. The LI-6800 interchangeable chambers allow the instrument to adapt as studies evolve. Collectively, the LI-600 and LI-6800 help students capture a more comprehensive dataset from their plants.
Additionally, the LI-600 is equipped with a Live View feature—when taking a measurement, data will appear in real time in the computer software and on any additional connected screen (e.g., a projector screen). This allows faculty to easily live stream data from the LI-600 in an educational setting. Simply attach the LI-600 onto a tripod via the tripod mount and let students view data right in the classroom. Similarly, the LI-6800 supports remote access, which allows faculty to view and control its console from an alternate source—such as a computer, tablet, or smart phone.
Each LEEF photosynthesis package is widely applicable to a variety of research applications and curriculums, including the following:
